人血管細(xì)胞間粘附分子-1(VCAM-1) 是一個由715個氨基酸組成的I型轉(zhuǎn)膜糖蛋白,分子量100到110KD,其分子內(nèi)還存在著七個C2型免疫球蛋白區(qū)。它的細(xì)胞外結(jié)構(gòu)是674個氨基酸多肽,隨后還有一個22個氨基酸的轉(zhuǎn)膜區(qū)和一個19個氨基酸的細(xì)胞內(nèi)結(jié)構(gòu)。在它的細(xì)胞外結(jié)構(gòu)存在著多個N-型糖基化位點(diǎn),因此它的細(xì)胞外機(jī)構(gòu)分子量可以增加到80KD左右(非糖基化分子量大約為74.14KD)。 此外, 每個C2型免疫球蛋白區(qū)之間是由一個二硫鍵所連接起來。人血管細(xì)胞間粘附分子-1和大、小鼠的種屬同源性在75%左右。但是,長度為19個氨基酸的細(xì)胞內(nèi)尾巴結(jié)構(gòu)在人、小鼠、大鼠之間是保守的。此外,小鼠表達(dá)的血管細(xì)胞間粘附分子-1,可以結(jié)合到小鼠和人的白細(xì)胞。說明小鼠和人的血管細(xì)胞間粘附分子-1具有類似的功能結(jié)構(gòu)區(qū)或同源性。研究資料顯示, 血管細(xì)胞間粘附分子-1也有分子變異現(xiàn)象,比如人的血管細(xì)胞間粘附分子-1可以有六個C2型免疫球蛋白區(qū), 而家兔可以發(fā)現(xiàn)8個C2型免疫球蛋白區(qū)的變異分子。
產(chǎn)品原理
本實(shí)驗(yàn)采用雙抗體夾心ELISA法??谷薞CAM-1單抗包被于酶標(biāo)板上,標(biāo)本和標(biāo)準(zhǔn)品中的VCAM-1會與單抗結(jié)合形成免疫復(fù)合物,游離的成分被洗去;加入生物素化的抗人VCAM-1抗體,生物素化抗人VCAM-1抗體與酶標(biāo)板上結(jié)合的標(biāo)準(zhǔn)品或樣本中的VCAM-1結(jié)合而形成免疫復(fù)合物,游離的成分被洗去;加入辣根過氧化物酶標(biāo)記的親合素,親合素與生物素特異性結(jié)合,游離的成分被洗去。加入顯色底物(顯色劑),若反應(yīng)孔中有VCAM-1,辣根過氧化物酶會使無色的顯色劑變藍(lán),加終止液變黃。在450nm處測OD值, VCAM-1濃度與OD450值之間呈正相關(guān),可通過繪制標(biāo)準(zhǔn)曲線求出標(biāo)本中VCAM-1濃度。
